Light micrograph of the leaf epidermis of the spring crocus Crocus vernus. The epidermis is very easy to peel off and is thus easily accessible for microscopic examination. In this preparation, the nuclei were stained with DAPI, a dye that specifically binds to DNA and RNA and makes the nuclei glow bright blue under UV excitation. In addition, red autofluorescence of the chlorophyll in the sphincter cells of the epidermis is seen. Microscopic contrast method : Fluorescence contrast with UV excitation.
